Ancient DNA Solves a 400-Year-Old Medici Murder Mystery — It Was Malaria, Not Poison
The deaths of two Medici brothers in 16th-century Florence have long been attributed to a family plot. New genetic analysis of their bones points to a far more mundane killer: malaria, carried by the same parasite that has claimed hundreds of millions of lives.

A new study took a different route. Rather than relying on historical accounts or chemical tests for toxins, the team turned to ancient DNA preserved inside the brothers’ bones. They sequenced material recovered from Francesco’s ribs and looked specifically for genetic traces of six Plasmodium species, the parasites responsible for malaria worldwide.
The result was clear. Malaria DNA was present, providing direct biological evidence that infection — not arsenic or aconite — brought the brothers down. Malaria was endemic in parts of Italy at the time, and a single severe bout could kill a healthy adult within days. The parasite leaves a signature in the skeleton’s DNA that can survive for centuries, turning a historical puzzle into a question of molecular biology.
The discovery does more than clear the Medici of one supposed crime. It also enriches the scientific record of malaria’s spread through the Mediterranean before the disease was finally eliminated from Europe during the twentieth century. A long-standing poison story, four hundred years in the telling, now gives way to the evidence preserved in bone.
